The tissueTUBEs TT05, TT05M, and TT1 are designed to withstand one impact. If a second impact is required, the extra thick (XT) version of each tissueTUBE is recommended. The extra thick bags can be impacted more than once. After the first impact, carefully inspect the tissueTUBE for punctures, re-freeze in liquid nitrogen, and impact again.
Multiple freeze/thaw cycles are never recommended for extraction of biomolecules. We recommend long-term storage of cryofractured tissues in small aliquots to avoid multiple freeze/thaw cycles.
No, the sample is not required to go through sample processing immediately. After cryofracturing, tissueTUBE plugs can be used to seal the tissueTUBEs for long-term storage at -80C, or vapor phase liquid nitrogen. Alternatively, the cryofractured tissue can be transferred into cryoTUBES for long-term storage.
