Covaris automation manager Kris Amirault presents a unified FFPE extraction library prep workflow designed to take samples from tissue to sequencing-ready libraries in two days, addressing the two most failure-prone stages of CGP testing. On the extraction side, the truXTRAC® SMART Solution on the Sonication STAR™ delivered consistently higher DNA purity and yield than a standard manual extraction method, directly translating to fewer re-extractions and reruns. Since implementing this FFPE extraction library prep workflow, Labcorp has achieved a 95% full report rate for OmniSeq INSIGHT samples, up from 82% with their previous method, and Covaris modeled the associated savings at approximately $1.3 million annually for a lab processing 10,000 samples per year.
On the library prep side, truCOVER library prep delivered unbiased fragmentation and uniform coverage across GC-variable genes, achieving F1 scores of 0.98 for SNPs and greater than 0.92 for INDELs from as little as 5 ng of FFPE-derived DNA, matching gold-standard genomic DNA performance. Together, the two workflows form a fully automated, high-throughput solution built for scalability, with continued development underway alongside Hamilton to further reduce hands-on time and implementation complexity.
