Title: Optimizing FFPE tissue stewardship: Covaris truXTRAC automated workflow delivers superior performance for tumor informed NGS assays

Description: This study presents data on the Covaris truXTRAC automated FFPE DNA and RNA co-extraction workflow with the Hamilton Sonication Star, a collaboration involving Covaris and Quest Diagnostics. The system demonstrated a ~3X increase in DNA yield per slide compared to current methods. This method was able to extract sufficient nucleic acid from challenging FFPE material, making it feasible to meet input requirements for multiple NGS based oncology assays using significantly less tissue. This automated approach enhances the tissue stewardship at large scale oncology diagnostics laboratories while also providing substantial cost benefits, including a reduced QNS rate, preventing repeat testing, and improving turnaround time.

Innovation Spotlight

Title: Optimized Automated Extraction for FFPE Tissue Cores

Description: Extraction of high-quality nucleic acids from formalin fixed and paraffin embedded (FFPE) tissue can be a time and labor-intensive process with limited through-put. Recent automation advances have allowed for the simultaneous extraction of high-quality DNA and RNA from FFPE tissue sections. Here we describe the validation and implementation of an optimized automation protocol using the Covaris truXTRAC FFPE extraction solution on the Hamilton Sonication Star for simultaneous extraction of nucleic acids from both FFPE tissue cores and sections. This modified protocol integrates Covaris’s Adaptive Focused Acoustics ® (AFA®) ultrasonication technology with Hamilton Robotics liquid handling to simultaneously provide NGS-quality DNA and RNA from both FFPE tissue sections and tissue cores.
