About this session:
Comprehensive oncology studies increasingly integrate liquid biopsy and archival tissue to capture complementary genomic and transcriptomic signals. NGS service labs often assemble extraction, library preparation, and enrichment kits from separate vendors to process plasma and tissue specimens, adding complex troubleshooting, validation burden, logistical overhead, and higher per-sample cost. This study tested whether a single-vendor, automation-ready Covaris workflow, spanning extraction through library preparation and enrichment, can serve as an end-to-end solution for matched plasma and FFPE samples.
Using 12 matched plasma and FFPE oncology samples, truXTRAC® FFPE extraction kits recovered gDNA and RNA from FFPE tissue, while the Apostle MiniMax® High Efficiency cfDNA Isolation Kit (Beckman Coulter) recovered cfDNA from matched plasma. Both cfDNA and gDNA underwent truCOVER® library prep, with truCOVER Whole Exome Kit enrichment generating exome, copy-number, and mitochondrial data; FFPE RNA underwent truCOVER® Total RNA Library Prep for RNA-seq. Performance measures include yield, coverage uniformity, on-target rate, duplication, concordance, and turnaround time, supporting flexible multi-omic endpoints for biomarker discovery and biopharma research.
